- Depression (ذہنی افسردگی) — treatment of major depressive disorder, including depression accompanied by anxiety, low mood, and loss of interest in daily activities

How Does Prothiaden 25mg Work?
Mood Stabilisation: Dosulepin blocks the reuptake of noradrenaline and serotonin — two key brain chemicals that regulate mood, motivation, and emotional wellbeing. By keeping these chemicals active in the brain for longer, Prothiaden gradually improves mood, reduces anxiety, and restores a more balanced emotional state. Full antidepressant benefit typically develops over 2 to 4 weeks of regular use — it is important to continue treatment even if improvement feels slow in the early days.
Pain Relief: Dosulepin also modifies pain signal pathways in the nervous system — reducing the intensity of chronic nerve pain independently of its antidepressant effect. This makes it useful for neuropathic pain conditions even in patients who are not depressed.
Sedative Effect: Prothiaden has a calming, sedative action that helps reduce agitation and improve sleep — making evening or bedtime dosing the most practical and comfortable approach for most patients.

Side Effects
Common: Drowsiness and sedation (most noticeable in early treatment), dry mouth, blurred vision, constipation, difficulty urinating, dizziness on standing up, weight gain, and increased sweating.
Less Common: Rapid or irregular heartbeat, tremor, confusion — particularly in elderly patients — and increased sensitivity of skin to sunlight.
Serious — stop and consult your doctor immediately if you notice: Chest pain or irregular heartbeat, seizures (fits), severe confusion or agitation, difficulty passing urine, or worsening of depression or new thoughts of self-harm — particularly in the early weeks of treatment.
Important: Antidepressants including Dosulepin may increase the risk of suicidal thoughts in younger adults during the initial weeks of treatment. Your doctor will monitor you closely during this period — always report any worsening of mood or distressing thoughts to your doctor immediately.
Who Should NOT Use Prothiaden 25mg?
- Known allergy to Dosulepin Hydrochloride or any other ingredient in the tablet
- Recent heart attack or any serious heart rhythm disorder
- Current use of MAO inhibitor medicines (MAOIs) — or within 14 days of stopping an MAOI — this combination is dangerous
- Severe liver disease
- Glaucoma (raised eye pressure) or urinary retention
- Children and adolescents under 18 years
- Pregnancy and breastfeeding — consult your doctor before use
