What is Nise 100mg Tablet? Nise 100mg contains Nimesulide — a preferential COX-2 inhibiting NSAID used for short-term management of acute pain, inflammation, and fever. Unlike traditional NSAIDs that equally block both COX-1 and COX-2 enzymes, Nimesulide preferentially targets COX-2 — the enzyme more directly responsible for inflammation — while relatively sparing COX-1, which protects the stomach lining. However, due to concerns about liver toxicity with prolonged use, Nimesulide is strictly recommended for short-term use only — generally not exceeding 15 days — and must always be taken under medical supervision with food.

How Does it Work? Nimesulide preferentially inhibits COX-2 enzyme — blocking prostaglandin production at sites of inflammation and in the hypothalamus where fever is regulated. By targeting COX-2 more selectively, it reduces pain and inflammation with comparatively less inhibition of COX-1 — the enzyme responsible for producing stomach-protecting prostaglandins. It also has additional anti-inflammatory properties beyond COX inhibition including inhibition of certain inflammatory mediators, contributing to its overall effectiveness.

Dosage and Administration

⚠️ Strictly short-term use only — do not exceed 15 consecutive days. Always take with food. Never combine with other NSAIDs, Aspirin above 75mg, or Paracetamol-containing products during Nimesulide therapy. Avoid alcohol completely during treatment.

Who Should NOT Take Nise 100mg?

  • Allergy to Nimesulide, Aspirin, or any other NSAID
  • Active stomach ulcer or history of gastrointestinal bleeding
  • Any liver disease or abnormal liver function — Nimesulide is contraindicated in all liver conditions
  • Severe kidney impairment
  • Severe heart failure or established cardiovascular disease

Side Effects

Common: Nausea, stomach discomfort, heartburn, mild dizziness, skin rash — reduced by always taking with food.

Serious — Stop and seek emergency help immediately: Yellowing of skin or eyes (یرقان — فوری ہسپتال جائیں), dark urine or pale stools indicating liver damage, severe upper abdominal pain, dark tarry stools or vomiting blood (پیٹ سے خون — فوری مدد لیں), severe allergic reaction with swelling of face or throat and difficulty breathing, significant reduction in urination indicating kidney problems, severe skin blistering or peeling.
